海水浸泡對表皮細胞凋亡和增殖的影響

摘要: 目的:探討海水浸泡對裸鼠表皮角質(zhì)細胞凋亡和增殖的影響及其意義。方法:24只雄性裸鼠隨機分為4組,應用光鏡、原位末端標記和免疫組化技術(shù),研究海水浸泡后裸鼠表皮角質(zhì)細胞凋亡和增殖的變化規(guī)律。結(jié)果:海水浸泡后,裸鼠表皮基底層和顆粒層角質(zhì)細胞發(fā)生凋亡,表現(xiàn)為細胞體積變小,細胞核固縮深染,細胞質(zhì)濃縮,嗜酸性增強,還可見核碎裂、溶解;原位末端標記顯示,對照組僅見基底層極少數(shù)陽性細胞,海水浸泡6和12 h,表皮各層見大量散在的陽性細胞,與對照組相比有顯著差異(P<0.01);Ki-67和細胞周期蛋白D1免疫組化結(jié)果顯示,對照組裸鼠僅基底層見少數(shù)細胞核呈弱陽性。海水浸泡3和6 h,Ki-67基底層陽性細胞輕度增加(P>0.05),海水浸泡12 h,基底層和顆粒層可見陽性細胞明顯增加(P<0.01);細胞周期蛋白D1表達增加在海水浸泡6和12 h組均有顯著差異(P<0.05或P<0.01)。上述改變均與浸泡時間呈正相關(guān)。結(jié)論:長時間海水浸泡可導致表皮角質(zhì)細胞的凋亡和增殖,兩者共同參與了海水浸泡對皮膚損傷的病理生理過程。
Objective: To investigate the effect of seawater(SW) immersion on the apoptosis and proliferation of epidermic keratinocytes in hairless mice skin.Methods: Twenty-four healthy male hairless mice were randomly assigned to 4 groups.the light microscope,TdT-mediated dUTP nick end labeling(TUNEL),and immunohistochemistry of Ki-67 and Cyclin D1 were used to study the apoptosis and proliferation characteristics of epidermic cells in the hairless mice skin following SW immersion.Results: Keratinocytes of both the basal and granulosum stratum showed apoptosis after SW immersion for 3 h,presenting cell shrinkage,condensation of nuclear chromatin,eosinophilic cytoplasm,and karyorrhexis or nuclear karyolysis,while in the control group,only low apoptosis was found by TUNEL in the basal stratum,and the apoptosis increased in the 6 h and 12 h SW immersion groups(P<0.01).The rate of Ki-67 positive cells increased slightly in the 3 h and 6 h SW immersion groups (P>0.05),but significantly in the 12 h(P< 0.01) group.Cyclin D1 expression was significantly up-regulated in the 6 h and 12 h groups(P<0.05,P<0.01).All the changes were positively correlated with the immersion time.Conclusion:Prolonged SW immersion can induce the apoptosis and proliferation of keratinocytes,both involved in the pathophysiological process.
